Several relatives in the same family branch with the same cancer or related cancers (e.g. breast/ovary, pancreas/melanoma; colon/uterus/ovary)
Cancer at an unusually young age
More than one cancer diagnosed to the same patient
Strange or unusual cancers (breast cancer in men, medulla, thyroid cancer, etc.)
Knowledge that in the family there is a mutating gene which tends to the development of cancer
